민아토 사카시는 천재적인 지능을 지녔지만, 압박감에 견디지 못해 대학 입시를 두 번이나 실패했다. 그 결과 많은 사람들로부터 바보와 실패자로 여겨지고 있다. 어머니의 또 다른 실망스러운 대화 후, 그는 공격을 받던 소녀 무즈비와 마주치게 된다. 무즈비는 도망치다가 민아토를 데려가고, 곧 민아토가 모르는 사이에 그는 '세키레이' 중 한 명인 '아시카리비'임을 알게 된다. 아시카리비는 유전적 특성으로 일부 '세키레이'와 파트너가 될 수 있는 신비로운 마스터 집단의 일원으로, 귀여운 소녀들, 풍만한 여성들, 그리고 미소년들로 구성된 108명의 세키레이들이 MBI라는 신비한 기업이 주최하는 경쟁에서 그들의 힘을 최대한 발휘할 수 있게 한다.

Minato Sahashi is extremely intelligent, yet due to his inability to cope under pressure has failed the college entrance exam twice. As a result he has been branded an idiot and loser by many. One day after talking to his yet again disappointed mother he comes into contact with a girl named Musubi who was under attack. The girl runs but takes him with her and soon finds that Minato, while unknown to him, is an Ashikabi, one of the mysterious set of masters that have the genetic trait that can become partners with some of the 108 cute girls, buxom women, and bishounen called "Sekirei". The reason for the partnership allows the Sekirei to use all their power to win in the competition held by the mysterious MBI corporation.
